La sindrome di Down (DO) è una sindrome genetica causata da un eccesso di materiale genetico sul cromosoma 21, che porta l’individuo ad avere tre cromosomi 21 invece di due. L’obiettivo generale di questo studio è stato quello di descrivere i fattori coinvolti nella predisposizione dei bambini con la sindrome di Down a sviluppare la leucemia megacarioblastica acuta (AML-M7). La metodologia utilizzata è una revisione della letteratura in formato narrativo. La SD presenta caratteristiche congenite come ritardo mentale, debolezza muscolare, anomalia cardiaca, tra le altre. Questi pazienti hanno anche un aumentato rischio di sviluppare la leucemia. D’altra parte, i bambini con SD hanno un rischio minore di sviluppare tumori solidi. Questa condizione leucemogena nella SD suscita interesse nello studio di queste patologie, poiché gli esperimenti suggeriscono che i bambini affetti da trisomia 21 hanno una probabilità 20 volte maggiore di sviluppare leucemie rispetto agli altri bambini, soprattutto LMA-M7. Studi genetici e molecolari hanno dimostrato che questa maggiore probabilità di sviluppare la leucemia è dovuta alla presenza di mutazioni somatiche nel gene GATA1.
