In den letzten Jahren hat sich die DNA-Sequenzierung der nächsten Generation zu einer neuen und leistungsstarken Technologie entwickelt, die einen viel umfassenderen Blick auf biologische Systeme auf genetischer Ebene und damit mehr medizinische Entdeckungen ermöglicht. Vor nicht allzu langer Zeit war die Sanger-Methode zur DNA-Sequenzierung, die als DNA-Sequenzierung der ersten Generation gilt, das Arbeitstier bei der Fertigstellung des Humangenomprojekts. Die Sanger-Methode war oft die beste Wahl für die De-novo-Sequenzierung, Resequenzierung, Genotypisierung und Fragmentanalyse. Die hohen Kosten und die geringe Geschwindigkeit der Sanger-Methode stellten jedoch eine Hürde dar, um im Zeitalter der personalisierten Medizin eine große Anzahl von Basen nach Bedarf zu sequenzieren. Um diese Nachteile anzugehen, haben viele Unternehmen und Forscher erfolgreich DNA-Sequenzierungsverfahren der 'nächsten Generation' entwickelt, d. h. jedes neue Sequenzierungsverfahren mit geringeren Kosten und höherer Geschwindigkeit, das nicht auf dem Sanger-Verfahren basiert. Dieses Kapitel wird zuerst die DNA-Sequenzierungstechnologie der nächsten Generation vorstellen und dann ihre Anwendung in medizinischen Entdeckungen zusammenfassen. Das Ziel dieses Kapitels ist es, die Leser mit der DNA-Sequenzierungstechnologie der nächsten Generation vertraut zu machen.
