Der systemische Lupus erythematodes (SLE) ist eine chronische Autoimmunerkrankung, die durch eine abnorme Produktion von autoreaktiven Antikörpern gekennzeichnet ist und zu einer Vielzahl von klinischen Symptomen führt. Natürliche Killerzellen (NK-Zellen), die drittgrößte Lymphozytenpopulation, tragen in hohem Maße zur Pathogenese des SLE bei. miRNAs sind kleine, nicht kodierende RNA-Moleküle, die über posttranskriptionelle Mechanismen eine wesentliche Rolle bei der Regulierung der NK-Genexpression spielen; dies wurde jedoch bei SLE bisher nicht untersucht. Ziel dieser Arbeit war es daher, die Expression von miRNAs zu untersuchen, die möglicherweise auf einen wesentlichen NK-aktivierenden Rezeptor, NKG2D, in peripheren mononukleären Blutzellen (PBMCs) und NK-Zellen von SLE-Patienten abzielen. Die beobachteten Anomalien in der Expression der Triade' miR-27a*, NKG2D und ULBP2' werden als entscheidende Aspekte bei der Entschlüsselung der beeinträchtigten Zytotoxizität der NK-Zellen von SLE-Patienten angesehen. Darüber hinaus konnte das miR-27a*-Imitat die Expression von NKG2D verstärken und könnte somit eine neue Grundlage für therapeutische Maßnahmen bieten, die auf NK-Zellen bei SLE-Patienten abzielen. Diese Arbeit wurde im Mai 2017 im International Journal of Rheumatic Diseases veröffentlicht.
