La fibrose pulmonaire idiopathique est une maladie chronique et progressive, caractérisée par une prolifération aberrante des fibroblastes et une accumulation des protéines de la matrice extracellulaire conduisant ainsi à une insuffisance respiratoire. Dans ce travail de recherche, notre choix s’est porté sur l’acide carnosique, substance bioactive extraite du romarin, pour évaluer son efficacité in vivo sur un modèle expérimental de fibrose pulmonaire induite par la bléomycine (BLM) chez le rat Wistar et in vitro sur les fibroblastes pulmonaires humains (HLF). La première partie in vitro a montré que l’AC induit l’apoptose des HLF d’une façon dose-dépendante, accompagnée par un arrêt du cycle cellulaire dans la phase G0/G1 et une activation de l’expression des protéines de signalisation Akt, p38-MAPK et p21. La 2ème partie, a confirmé l’effet protecteur de cette biomolécule in vivo sur le processus fibrosant et le stress oxydant induits par la BLM. A la lumière de ces résultats, il serait intéressant de tester l’ efficacité de l’AC sur certaines maladies dont la physiopathologie est dominée par l’inflammation, le stress oxydant et la prolifération cellulaire.
