Lo scopo di questa indagine è stato quello di sviluppare e ottimizzare la claritromicina caricata con acetato di pullulano per un’applicazione a rilascio prolungato mediante la metodologia della superficie di risposta basata sul disegno fattoriale con modello 2FI ridotto. Vari parametri come RPM, tempo, concentrazione di farmaco (Claritromicina) e concentrazione di acetato di pullulano sono stati ottimizzati rispetto a F5. Si è riscontrato che RPM e tempo avevano un effetto maggiore rispetto alla concentrazione di claritromicina e di acetato di pullulano per il rilascio prolungato del farmaco. È stato tracciato un grafico di risposta 3D e sono state selezionate le interazioni ottimali mediante ricerca di fattibilità e griglia. Le risposte osservate coincidevano bene con i valori previsti dal disegno sperimentale. La formulazione ottimizzata F5 ha mostrato un rilascio prolungato e sostenuto di claritromicina per 7 ore. Il profilo di rilascio della microsfera caricata con il farmaco rivela che il pH del mezzo di coltura lo influenzava in condizioni in vitro. Nelle condizioni ottimizzate, il rilascio prolungato può essere raggiunto fino a 9 ore. Pertanto, l’acetato di pullulano caricato con claritromicina è un materiale polimerizzato utile per lo sviluppo e la formulazione di farmaci sensibili al pH.
