O sistema de distribuição de fármacos gastro-retensivos pode reter o medicamento na região gástrica durante várias horas, o que melhora a libertação de fármacos e melhora a biodisponibilidade. Atenolol, um fármaco anti-hipertensivo tem meia-vida curta, biodisponibilidade limitada, altamente instável a pH básico e é extensivamente absorvido do estômago. Estas características despoletaram a necessidade de desenvolver um sistema gastroretentivo. No presente estudo foi desenvolvido um hidrogel superporoso como um sistema de fornecimento de fármacos gastroretentivos. Hidrogel superporoso do tipo rede de polímeros interpenetrizados Chitosan / poli (álcool vinílico) foram preparados utilizando um método de espumação de gás utilizando o glioxal como agente reticulado. Os comprimidos de atenololol SPH foram preparados por compressão directa incorporando celulose microcristalina, estearato de magnésio, talco e bicarbonato de sódio, etc., e avaliados. Os hidrogéis superporosos eram altamente sensíveis ao pH dos meios de inchaço e apresentavam um inchaço rápido e boa porosidade. Os comprimidos de atenolol SPH mostraram boas propriedades pré-compressivas e pós-compressivas. A formulação IV exibiu uma boa taxa de inchaço e libertação sustentada de droga durante o período de 12 h através da gastro-retenção.
