VV (Vaccinia-Virus) ist eines der komplexesten Viren mit einer Größe von mehr als 300 nm und mehr als 100 Strukturproteinen. Sein Zusammenbau beinhaltet aufeinanderfolgende Interaktionen und größere Umlagerungen seiner strukturellen Komponenten. In dieser Studie wurden infizierte Zellen mit Hilfe der Lichtfluoreszenzmikroskopie selektiert und anschließend mit Hilfe der Röntgenmikroskopie unter kryogenen Bedingungen abgebildet. Tomographische Kippserien von Röntgenbildern wurden verwendet, um dreidimensionale Rekonstruktionen zu erstellen, die verschiedene zelluläre Organellen (Kerne, Mitochondrien, ER) zusammen mit zwei anderen Arten von Viruspartikeln zeigen, die mit verschiedenen Stadien der Vaccinia-Virus-Reifung (IV) unreife und (MV) reife Partikel verbunden sind; Witferin-Assays zeigten eine Bindung an Aktin, die die Polymerisation und Dehnung der Filamente verhindert und zu falsch verpackten oder abweichenden Virionen führt, was das Fortschreiten der viralen Infektion hemmt. Die Ergebnisse zeigen, dass die Röntgenkryotomographie ein leistungsfähiges Instrument zur Erfassung dreidimensionaler Strukturinformationen aus gefrorenen, unfixierten ganzen Zellen ist.
